Effects of Non-Essential Amino Acids on Knee Joint Conditions in Adults: A Randomised, Double-Blind, Placebo-Controlled Trial

<Background>
 Joints act to absorb the physical shocks that occur during physical activity. Therefore, joint problems reduce the performance of exercise and daily activities, affecting the quality of life. Therefore, in this study, we decided to verify whether six non-essential amino acids (serine, alanine, glycine, glutamic acid, sodium aspartate, and proline: hereinafter referred to as a six-amino acid mix) are useful for improving the condition of human knee joints.

Methods
 The study was designed as a placebo-controlled, randomized, double-blind, parallel-group comparative study, with 50 adult men and women. Participants were recruited from 0 to 20 years old who had knee joint discomfort but had not been diagnosed with joint disorders (Kellgren-Laurence classification 64 or I). The test foods were a control food containing no amino acids or a "6-kind amino acid mix," and participants were asked to take 1g of the food three times a day for 4 weeks.

 The primary outcome measures were the visual analogue scale (VAS), the Japanese Knee Osteoarthritis Measure (JKOM), and the Japanese Orthopaedic Association (JOA) score, which were used to evaluate pain, stiffness, and discomfort in the knee. Each outcome measure was measured before, 4 weeks after, and 12 weeks after the start of the test food intake.

<Result>
 The group that consumed the "6 Amino Acid Mix" showed a significant improvement in knee joint pain, stiffness, and discomfort compared to the group that consumed the control food (Figure 1). Similarly, in the JKOM and JOA groups, the group that consumed the "6 Amino Acid Mix" showed a significant improvement (Figures 2 and 3).

<Conclusion>
 The 6 Amino Acid Mix has been shown to improve everyday movement, joint pain, stiffness, and discomfort in people with joint discomfort.
